Many good opinions here.  But, I believe Dwight Yoakum has clearly had the most success as a musician transitioning to serious acting.  He has had more legitimate acting roles with real substance, and in some very good movies, than pretty much anyone else mentioned.  I've been quite amazed with his acting abilities since Sling Blade.  He's the real deal.

The lines get a little fuzzy for some of them, as, for example, Jamie Foxx and Russell Crowe are successful musicians who are more famous as actors.  That I haven't seen mentioned yet: Courtney Love was good in The People vs Larry Flint.  Puff Daddy was pretty damn funny in Get Him to the Greek.  Many people probably know Kris Kristofferson and Jerry Reed more for movies than music.
