I am assembling my own upper and need to know how/if I should check headspace?

Headspace is set at the factory when the barrel extension is affixed.

No, but if it gives you peace of mind, then by all means do it.  Personally, with your the barrel being a Noveske I wouldn't worry about it.

Checking headspace is a basic safety measure. Especially if any of the parts are used.
It is unlikely that a new bolt and new barrel would be unsafe to fire, but headspace varies when mixing and matching parts.
Since headspace is measured from the face of the bolt the true measurement must be made with the bolt the barrel will be used with.
Personally I like my new weapons to start out at minimal headspace. YMMV
