Just got a Burris Fastfire and mount for my 22/45 handgun.

How is the stock/original rear sight removed to mount the Fastfire? Does it need to drifted or tapped out? I don't see how it could just be unscrewed or otherwise....

Thanks!

Did your 22/45 come with mountable "rail?"

If it did; wouldn't the fastfire just mount to  that?

Yep - if your 22/45 is tapped for a scope rail, get that and use the Weaver base that came with the Fastfire to mount it.  This is going to mount the sight a little higher, but it will leave you the option of returning to your open sights if you want.

Otherwise, you have to drift the rear sight out of the dovetail.  You then drift in the Fastfire mounting dovetail and the rest of the mount screws to that.  I just use a brass punch and a mallet to drift the rear sights.
