Does extra room for the powder in the case degrade accuracy?

Too many it depends to give a straight answer.

What cartridge, powder, bullet, and firearm?

Generally it's best to have an almost case full of powder, but there are a lot of exceptions.

+1

As a general rule a high load desity is prefered for accuracy.

300 win mag. Case being loaded to spec from spear loading 13th edition. Seems like Re15 only fills like 75% of case leaving room for the powder to move a good bit.

If you are using a listed load from a reloading manual, you will be GTG. As you are.

Reason why the case isn't full is that Re-15 is a "fast" burning powder when used in 300 Win Mag.

In 223, Re-15 is a "slow" burning powder and normally compressed. Case size is different between these two.

Try some IMR-4831 or H-4831 as both of these are slower burning and will give better case fill.

There are light .38 Special loads that should not be used in a .357 case.

Generally is the operative word here. By the time you stuff a pill in there that case volume will quickly disappear.
Your good to go.

You'll usually get a higher vel and better accuracy with a case full of slower powder, like 4831 in a 300.
RE15 is usable in the 300, but vel is low.
