What markings should be found on the Colt detachable handle?
I have a Colt LE6920.  Are all the handles off new Colts the same?
Are the Colt Defense and Colt Manufacturing handles the same?

No, they're not the same.  Some have what looks like a cardinal head, some have what looks like a keyhole marking - sometimes followed by a number or letter, some have a broken "A" - sometimes followed by a number or a letter and some have nothing at all.

I don't know about the rest of your question, Colt Defense vs. Colt Manufacturing, though.

I have one from a Colt M4 (LE model) and it's marked with a broken 'A' which is just a forging mark which means Anchor Harvey Aluminum forging, and a letter 'B' on the left side of the handle.

The markings will vary because they get their forging from different sources.

The markings could also be located on the right or left side.

Colt Defense and Manufacturing are the same parts with the noted differences in markings.

Mine has the #10 and the keyhole.
Does the keyhole have any meaning?

It's the mark of the company that supplied Colt the raw forging, which Colt finished.  That company is Cerro Fabricated Products.

This one shows the marking by the rear adjustable sight pretty well
