Urban home defense.  Homes are typical wood construction.  No suppressor.

Do you go with:

a.) .300 Blackout in a 10.5" barrel slinging 110gr Barnes VOR-TX (aka "Black Tips") or 110gr Hornady V-MAX
    Pros: typically more than sufficient penetration at around 16"+ in gel, powder burns in about 9" of barrel so should have less blast than 5.56.
    Con: often penetrates all the way through deer and the like.  Maybe too much penetration?

or

b.) 5.56 NATO in a 11.5" barrel slinging 77gr Black Hills Tipped Match Kings
    Pro: less penetration at about 14'-16" in gel,
    Con: maybe insufficient penetration on Fat Bastard and blasty out of a pistol / SBR barrel.

5.56 and I wouldn't even bother with exotic ammo, just m193 or some kind of hunting ammo.
Don't over think it.
5.56 from a 10.3" is good enough for Navy seals.

If I could get 300BLK in quantity and price of 5.56, I would.

From an 11.5" gun, using any Barnes TSX will probably be effective.

I was going to say I voted for .223 but all my ammo is 55 grain FMJ.  

I've got some heavy stuff in the garage waiting to be reloaded/tested for accuracy this winter, but it's not for regular use.  Don't need to shoot 600 yds. around here when 100 yds. is about as far as you can see.

Home defense includes civil insurrection, riots, looters and even warfare and terrorist attacks. Not just simple home invasions, burglary or robbery.

Having a rifle that shoots the same ammo as the local law enforcement and the military is critical to survival.

It'll work fine for close quarters, even in short barrels. I prefer CBC/Magtech Mk262, but any 5.56 will put dicks in the dirt at close range.

This is really best answered with "both".  For home defense, I would really want to suppress my SBR (or "pistol") and .300BO is more effective in that department.  However, for training, 5.56 is far cheaper.  I think both are effective enough and I really don't think there would be any drastic change in terminal ballistics 50 meters on in (maybe out to 100 meters?).  A duplicate upper, on in 300 and one in 5.56 would all a better performing, suppressed choice with 300BO and a far cheaper 5.56 choice for training.  If you're just getting the one, I would go with 5.56, but I would still want to suppress that beast indoors!

Why?  Do you plan on raiding their armories?  They certainly won't be handing out ammo to civilians

Would agree, with one BIG caveat ... that being, any 5.56 SBR you might shoot inside a structure needs to be suppressed, or you and anyone nearby WILL incur significant auditory damage from the blast of rounds.

any 300BO needs a can as well, its not quiet, if your arguing running subs to make it quiet but give up all the ballistics of supers-well then you have a fine day

Either way the gun would be suppressed. I have a 8.2” 300 blk with suppressor and a 10.5” 5.56 with suppressor. I’d take the 5.56 because it has a 1-8 scope on it.
