I don't know for a fact that Alexander was, but it was definitely a very practiced thing back then.

Alexander married Roxane, a Persian princess. He later took a second wife named Statira (murdered by Roxane after Alexander's death). Roxane also bore Alexander a son (Roxane and her son were murdered by Cassander in 310 B.C.). These are
facts. There is no question. Rumors of a homosexual relationship with Hephaistion are completely unsubstantiated and will remain a historical mystery until someone invents a time machine.
Alexander's marriages to Roxane and Statira are unquestioned. Any "funny" business is conjecture. Therefore, I don't think it's appropriate to call him "queer" with no more evidence than what amounts to rumor and speculation.
That said, it wouldn't change my opinion of him if historians somehow discovered he
was bisexual. You are correct that homosexuality certainly was not frowned upon by the ancient Greeks and Macedonians as it is in our society and this is no doubt one of the reasons such rumors have always been a part of Alexander's legend.
Oh, and Oliver Stone is a fucking idiot.
