We (two couples) are looking to visit a winery in that SW Michigan area. Any suggestions, especially since we would be remaining overnight?

If you want wine, come over the border into southwest Michigan, about another 20 miles.  There's Tabor Hill, Gravity, Round Barn, Chill Hill, Domaine Berrien, Lemon Creek, Free Run, St Julian, Red Top, Founders, and Dablon to name a few.

Tabor Hill and Round Barn Public House serve food.  For beer there's Greenbush, Haymarket, Tapistry, Watermark,  and Round Barn Public House.  The brew places serve food.

All these are fairly close within proximity of each other, and there are "wine trail" signs you can follow.  Head towards the town called Baroda.  Everything is within proximity of it.

Are there any preferences toward hotels with centralized location, accommodations, etc?

The hotels in Stevensville that are close to I-94 and fairly modern.

Moondog pretty much nailed it.  Haymarket has excellent beer and the wineries are fairly close together.  Grab a wine trail map from a google search.
