To qualify the line must be the last(or close to last) spoken in the movie, and must serve as a punchline to the whole thing.

Best example:

Stalag 17, "Maybe he just wanted to steal our wire cutters. You ever think of that?"

Another example:

The Big Lebowski, "Fuck it dude, let's bowl."   Technically, this isn't the last line in the movie as the stranger still has his ending monologue. But as part of the film narrative, this closes it out and serves the purpose.

Another example:

Bladerunner, "It's a shame she won't live, but then again who does?"   Again, this is problematic in it's a repeat of lines spoken earlier and Deckard is only remembering them.   Also problematic in that this is the "Final Cut" version with the paper unicorn smashing and all the discussion that brings up.

So...  What's your favorite final punchline to a movie.   I think the Stalag 17 line is the perfect example and best way to finish that movie.

What else is there?

Edit - Not a punchline, but I use it as one!

That’s a very under-rated movie. Watched it with my step-kids a few years ago and we all really enjoyed it.  Good call.

When some wild-eyed, eight-foot-tall maniac grabs your neck, taps the back of your favorite head up against the barroom wall, looks you crooked in the eye and asks you if ya paid your dues, you just stare that big sucker right back in the eye, and you remember what ol' Jack Burton always says at a time like that: "Have ya paid your dues, Jack?" "Yessir, the check is in the mail."

Technically, the last line was "What was that?" Excellent punchline.
